I've always wondered what was the story behind that remark. By the way Elvis turns to the backup singers and smiles after having said that, one would guess it's some kind of inside joke. Just wish we could know what the joke was.

There is no 'story' behind the remark. Elvis says as quoted above before the end of a song after hearing screams from pulling up his belt. The Sweet Inspiration's burst out laughing as its typical funny Elvis behavior, he grins and throws them a punch.
